This is an unusual tetrazine that displays low reactivity in IEDDA reactions due to the steric hindrance of the bulky tert-butyl substituent. Nevertheless, it has two notable applications: 1) Bioorthogonal activation of fluorophores via an isonitrile–tetrazine click-to-release reaction; 2) Use as a bioorthogonally removable protecting group, cleavable by isonitriles.
